Characterization of circulating insulin and proinsulin-binding antibodies in autoimmune hypoglycemia.
Five patients with fasting and(or) postprandial hypoglycemia were found to have insulin antibodies in the absence of previously documented immunization. Studies on the equilibrium-binding of insulin to the autoantibodies revealed two classes of binding sites with association constants and binding capacities analogous to those of insulin antibodies from insulin-treated diabetic patients. Similar...
متن کاملInsulin-binding antibodies in relation to insulin therapy.
Methods for detecting insulin binding or transporting antibodies are described, involving the use of radioactively labelled insulin (insulin (131)I). These antibodies are detected in a high proportion of patients receiving insulin, particularly those requiring large doses. The protein fractions concerned appear to be the beta and gamma globulins. The significance of the phenomenon is discussed.
متن کاملMonoclonal antibodies to the human insulin receptor block insulin binding and inhibit insulin action.
Antibodies to the insulin receptor were prepared in BALB/c mice by immunization with IM-9 human lymphocytes, a cell type that has a large number of plasma membrane insulin receptors. The spleens of these mice were then removed, and their lymphocytes were fused to a mouse myeloma cell line, FO cells. After screening over 1,200 resulting hybrids, one stable hybrid was obtained that produced IgG1 ...
متن کاملEquilibrium binding studies of insulin antibodies in diabetic subjects.
Equilibrium binding of insulin to human insulin antibodies from diabetic subjects has been studied by two methods. Results obtained by using a nondissociating gel filtration assay differed from those obtained by a conventional competitive binding method. Both methods yield data consistent with two classes of insulin binding site, having association constants of approximately 10(9) and 10(7) lit...
متن کاملThe subunit structure of rat liver insulin receptor. Antibodies directed against the insulin-binding subunit.
A highly purified preparation of rat liver insulin receptor obtained by affinity chromatography has two main peptide components with M, values of approximately 135,000 and 45,000. Peptide maps of the two components after digestion with chymotrypsin indicate the 45,000-dalton component i s not a fragment of the 135,000-dalton component. Antibodies prepared against the 135,000-dalton component i...
